GENETIC EFFECTS IN SOMATIC AND GERM CELLS IN RABBITS FOLLOWING EXTERNAL GAMMA RADIATION II. Chromosome aberrations in peripheral blood lymphocytes

چکیده
The aim of the present study was to evaluate the effect of gamma irradiation on the hereditary structures of peripheral blood lymphocytes and to quantify this in a dose-effect study. We used adult male rabbits subjected to total gamma irradiation at a dose range of 0.5 − 3.0 Gy. Twenty-four (24) hours later, we began the process of culturing and determination of chromosome aberration frequency using peripheral blood lymphocytes from the blood of these animals. The analysis of results showed a dose-dependent increase in chromosome aberration frequency. The highest percentage of aberrant cells (29.3±2.02) was observed following irradiation at 3.0 Gy. The regression analysis of results showed that the dose-effect relationship (yield of dicentrics, fragments and percentage of abnormal cells) satisfied the function, y=αD.

Background: Sperm chromatin insufficiencies leading to low sperm count and quality, infertility and transmission of chromosomal microdeletion and aneuploidies to next generations can be due to exposure to environmental pollutions, chemicals and natural or manmade ionizing radiation. Male rabbits, aged 4.5 months, were subjected to graded total gamma irradiations of 0.5 Gy, 1.5 Gy, 2.5 Gy and 3.0 Gy with a dose density of 24 Gy/min. The genetic effects in spermatogonia were assessed 3 months later using cytogenetic analysis of chromosomal preparations. The data showed a dose-dependent increase in the frequency of reciprocal translocations, induced in spermatogonia and detec...
متن کاملFuture research directions to study genetic damage in germ cells and estimate genetic risk.
The late Frits Sobels developed a parallelogram model to estimate genetic risk to humans based on experimental data in somatic cells (peripheral blood) of exposed animals and humans and on data from progeny studies of exposed animals (mice). Recently, an extension to the original parallelogram model was proposed to bridge the gap of extrapolation between rodent and human germ cells by studying ...

Animals were sacrificed at 6, 12, 24 and 48 weeks after the start of the exposure for analyses of reciprocal translocations in peripheral blood lymphocytes and germ cells. The frequency of the total chromosomal aberrations in the peripheral blo...
